Description:

The Mokuton bloodline limit ability consists of creating wood by the combination of earth and water chakra elements. This includes the spontaneous growth of trees and the use of pillars of wood as weapons or to build structures. The Mokuton, like other charka fusion techniques such as the Hyoton, requires the perfect control of two specific chakra elements in order to merge them.  Only a person with this particular Kekkei Genkai can accomplish this.  As with most jutsus, hand seals are still required to perform Mokuton.

The original user of Mokuton was Hashirama Senju who became the First Hokage of Konoha. He used it not only in battle but also to aid the initial construction of Konoha. He performed his Mokuton Hijutsu: Jukai Kotan  (Wood Release Secret Technique: Birth of Dense Woodland) to grow the thick forests that surround the Hidden Village in the Leaf (in the anime).  Because of his unique abilities, Hashirama was named the First Hokage, and won over Madara Uchiha, the leader of the Uchiha clan at the time.

Surprisingly, none of Hashirama’s descendants can use the Mokuton ability. It is unknown whether it is because they did not inherit the Kekkei Genkai or if they simply were unable to awaken it.  The Mokuton was unique to Hashirama Senju until Orochimaru extracted his DNA from his remains and injected it into 60 test subjects, all of them young children.  Nearly every child died in reaction to the foreign DNA, forcing Orochimaru to abandon the experiment.  But unbeknownst to him, one child survived and inherited Hashirama’s Kekkei Genkai.  He would grow up to become a member of the Leaf’s ANBU named Tenzo, better known by his codename Yamato by the other members of Team Kakashi.

Hashirama also mastered the ability to control the bijuu or tailed beasts.  Yamato also possesses this skill to some degree, and is able to suppress the Kyuubi’s chakra inside Naruto.  Yamato used this power to revert Naruto back from his four-tails transformed state when he used the sealing technique Hokage Shiki Jijun Jutsu: Kakuan Nitten Suishu (Hokage-Style Sixty-year-old Technique: Enclosed Hermitage Return to Society) to create wooden dragons which attach to him and force the Kyuubi's chakra to recede.

It is possible that Hashirama’s necklace, which Tsunade (his granddaughter) gave to Naruto, aided to amplify the power of Yamato’s ability. But yet another possible theory suggests that the necklace’s purpose is to prevent the Sharingan from gaining control of the Kyuubi.
